If a merchant charged your card and has taken money from it when they shouldn't have or you purchased something online and didn't receive the purchase then yes you can put in a dispute with the bank and merchant on the charge.

If the bank finds enough proof that you should be refunded then they will do a chargeback which forcefully reverses the transaction and returns the funds to your bank account.

You must have enough proof etc to prove you didn't get the item you purchased or that you cancelled something and they still charged your card anyway.

First contact the merchant though and work things out with the merchant because most merchants that are legit will be glad to refund you without you having to go into a dispute on the debit card because when a chargeback occurs the merchant is charged around a $15.00 to $25.00 chargeback fee which adds up in costs for the merchant.
