Tamiflu can take up to 48 hours to take effect so you should take the Tamiflu 48 hours before the onset of severe symptoms.
Tamiflu doesn't actually treat or get rid of the flu it just makes the symptoms less severe if taken soon enough.
If you wait to take it 48 hours later then the Tamiflu won't do any good.
