Most vehicles will require you to check the transmission fluid warm while the engine is running and the transmission is either in park or neutral.
It should say on the transmission fluid dipstick whether it needs to be in park or neutral to check the transmission fluid.
If you have to check the transmission fluid in neutral be sure to apply the parking brake to keep the vehicle from running you over should it move.
