It really makes no difference in whether you turn the desktop computer off at night or leave it on in sleep mode.
I personally just put my desktop computer in sleep mode overnight and haven't had any problems with the computer.
If I'm gonna be gone away from my house and won't be using the computer longer than one day then I shut it completely down and unplug it just in case lightning were to strike or a power surge happens when I'm gone.
